The measurement of the electrophoretic mobility of proteins by microelectrophoresis using up to date, commercially available, instrumentation is discussed. The primary systematic uncertainties and their effects such as joule heating and convection, the presence of aggregates and polarisation concentration are discussed along with methods for their detection, quantification and reduction. Simple measurement strategies are found and excellent agreement between microelectrophoresis and literature values from other techniques is demonstrated.
