Abstract
⺠Twelve enzymes expressed and purified for reconstituting fermentative glycolysis. ⺠Simple assays are described for each glycolytic enzyme. ⺠In vitro fermentation in isolation recycled ATP with glucose as substrate. ⺠The ratio of glucokinase to phosphofructokinase was critical for this process. ⺠Engineered pyruvate decarboxylase allowed faster ethanol production.
