Abstract
Peroxisome proliferators activated receptor gamma (PPARγ) plays a rather crucial role in many human chronic diseases. In this study, a biochromatography model based on M1.3 origami-PPARγ with offline HPLC/MS was built, as an efficient, rapid, cost-effective drug screening system, it was applied to separate and identify the potential active ingredients that act on PPARγ from Ginsenosides. M1.3 was excised from self-produced M13mp18ssDNA, then it was folded into M1.3 origami with 24 staple strands. PPARγ-M1.3 origami conjugates bonded on the surface of silica was used for the preparation of biochromatography stationary phase. The fractions retained by M1.3 origami-PPARγ column were collected and identified by HPLC/MS system. By this biochromatography model, Ginsenoside Re was identified as the potential active ingredients from Ginsenosides. Therefore, this biochromatography model can be applied for screening active components acting on PPARγ from traditional Chinese medicines.
