► C16-Cer increases mouse ESC migration. ► C16-Cer regulates PKC and FAK/Paxillin-dependent Cdc42 activation. ► C16-Cer treatment leads to formation of N-WASP/Cdc42/Arp2/3 complex. ► F-actin expression is increased following C16-Cer treatment. ► C16-Cer promotes the interaction between cofilin-1 or α-actinin-1/-4 and F-actin.
