Abstract
Expansion of the acetonitrile-assisted, diphosgene-mediated synthesis of bicyclic pyrimidines (compound B) expeditiously afforded dual orexin receptor antagonists (DORAs) with excellent potency and rat sleep efficacy. In vitro bioactivation studies on key compounds are also described.
