Abstract
The mean molecular weight of synthetic oral drugs has increased substantially over the past 60 years. Fewer than 5% of oral drugs approved or marketed since 1937 have more than 4-H bond donors and only 2% have MWÂ >Â 500 and >3 H-bond donors.
