Abstract
► We characterized a midgut-specific chitin synthase gene (LmCHS2) from Locusta migratoria. ► LmCHS2 was mainly expressed in apical regions of brush border forming columnar cells of gastric caeca. ► RNAi of LmCHS2 led to a feeding cessation and high mortality of the insect. ► RNAi effect attributed to a reduced synthesis of chitin and disruption of peritrophic matrix (PM) formation. ► LmCHS2 is responsible for the biosynthesis of chitin associated with PM and plays an essential role in locust growth and development.
