Abstract
► Genome is constantly exposed to various endogenous and exogenous agents that cause DNA double strand breaks (DSBs). ► DSBs, if left unrepaired, cause genome instability and cancer. ► The ubiquitin-proteasome system is virtually involved in any cellular process. ► By summarizing the current literature we observed that the UPS is an integral part of DSB repair. ► Based on this, we propose new perspectives for anti-cancer therapy.
