Abstract
► Mathematical models allow us to test hypotheses about stem cell behavior. ► We model homeostasis and monoclonal conversion in a colonic crypt. ► Asymmetric division without feedback cannot produce a stable structured crypt. ► Under symmetric division within a stem cell niche, a stable crypt arises naturally. ► The spatial structure of a crypt significantly affects the time to monoclonality.
