Abstract
⺠Mathematical models allow us to test hypotheses about stem cell behavior. ⺠We model homeostasis and monoclonal conversion in a colonic crypt. ⺠Asymmetric division without feedback cannot produce a stable structured crypt. ⺠Under symmetric division within a stem cell niche, a stable crypt arises naturally. ⺠The spatial structure of a crypt significantly affects the time to monoclonality.
