► In silico analysis of posttranslational modifications of occludin have been reported. ► Several novel phosphorylation and O-glycosylation sites have been identified. ► Activity of ERK and GSK-3 kinases have been proposed in occludin. ► Phosphorylation and O-glycosylation interplay as mechanism to impair HCV entry is reported.
