Abstract
The enhanced production, purification and applications of xylanase from a mixed microbial culture namely Acetobacter xylinum NCIM 2526 and Cellulomonas uda NCIM 2523 using Prosopis juliflora was studied. Significant medium components for xylanase production were optimized using a statistical method, Central Composite Design (CCD) in a shake flask culture. An optimized xylanase activity of 25.32Â IU/mL was achieved after the statistical experiments and this is nearly 5 fold more than the unoptimized value, 5.85Â IU/mL. The produced xylanase was partially purified using salting out, dialysis and gel filtration chromatography techniques. Two homomers of xylanase were recovered with molecular weights 133.52 and 42.66Â kDa. The partially purified enzyme was tested for its potency to synthesize xylooligosaccharides (XOS) from the beech wood xylan. Further, in-vitro prebiotic activity of XOS with probiotic organisms was determined. XOS at 2% w/v was favored the growth of all the tested Lactobacillus strains.
