Abstract
An efficient laccase production fungus Trametes sp. LS-10C was isolated and identified, and the laccase producing medium was optimized by response surface methodology (RSM) in shake flask fermentation. The highest laccase activity obtained at 9 d in shake flask and 10-L fermentator were 588.61±2.2 U mLâ1 and 873.82±22.91 U mLâ1, respectively. The molecular weight of purified laccase determined by SDS-PAGE was around 65 kDa. The optimal pH was 4.0-4.5 and the optimal temperature was 40 °C. Among the tested substrates of ABTS, DMP and guaiacol, both the highest values of Vmax (11.4±1.5 mM minâ1) and kcat/Km (6653.2±18.2 mMâ1 sâ1) were found for ABTS and the lowest Km value (38.1±1.7 µm) was found for DMP.
