► Parameters not always considered in many germination protocols may affect counts. ► Counts are increased by initial hydration status of conidia. ► Counts are increased by immersion time and incubation time. ► Use of stains and/or coverslips may profoundly affect viability assessment. ► Counts for high-quality conidial batches are not affected by the previous parameters.
