•Effectively concentrating bacteria by MNP based immunoassay using microfluidics.•Microchannels are fabricated easily and reproducibly in one step.•Different concentrations of Escherichia coli were detected with high degree of repeatability.•Inexpensive yet powerful tool to quantify bacteria is demonstrated.
Here we report development of a magnetic nanoparticles based immunosensor chip comprising of specially designed circular microchannel and embedded miniature permanent magnet. The present process of fabrication allows a rapid and reliable tool to create microchannels with ease and at a low cost. The chip permits easy in-flow capture of bacteria, their visual detection and quantification as demonstrated by Escherichia coli detection. Such a technique enhances capture efficiency and makes quantification of captured antigens simple.
