► Long-term treatment with l-isoleucine or l-leucine in AIN-93G diet has promoting effects on rat bladder carcinogenesis. ► For the same treatment, effects of l-isoleucine or l-leucine in MF diet were not obvious. ► Inconsistent effects of l-isoleucine or l-leucine might partly due to polyphenols contained or not in the basal diet. ► Urinary pH value and urine sediments are not related with mechanisms of their promoting activities.
