Abstract

Rat myocardiocytes H9c2 were pre-treated with rice protein hydrolysate and then treated with hydrogen peroxide (H2O2). The apoptosis rate and caspase-3 activity of H9c2 cells were analyzed by Flow Cytometry and Caspase Activity Detection Assay, respectively; the expressions of B cell lymphoma/leukemia-2 (Bcl-2) and Bcl-2 associated X protein (Bax) were determined by real time-PCR and Western blot. The results showed that apoptosis of H9c2 cells was alleviated in response to the treatment of rice protein hydrolysate, potentially resulting from the up-regulation of Bcl-2 and the down-regulation of Bax. The results indicated that rice protein hydrolysate attenuated the apoptosis of myocardiocytes H9c2 involving in the Bcl-2/Bax pathway during H2O2 challenge, which suggest a potential role of rice protein hydrolysate on the healthcare of cardiovascular disease, especially acute myocardial ischemia.

► Anti-apoptotic effect of rice protein hydrolysate on H9c2 was determined. ► The apoptotic changes of H9c2 were determined. ► Rice protein hydrolysate attenuated the H9c2 apoptosis through bcl-2/bax pathway. ► Mechanism of rice protein hydrolysate against apoptosis in H9c2 was investigated.
