► LmDHODH crystal structures have been solved in complex with orotate and fumarate. ► Orotate and fumarate have been found to occupy the same active site. ► A cation ion has been found in a cavity located at the dimer interface. ► Dimer interface have direct influence on the conformation of the catalytic loop. ► Trypanosomatid DHODHs share high level of sequence and structural similarity.
