► Surface plasmon resonance was used to survey myosin assembly. ► Four positive residues guiding myosin polymerization in vitro are defined. ► Their topology in the myosin coiled-coil rod is unique. ► Their mutagenesis affects myosin incorporation in the sarcomere in vivo.
