► We have demonstrated that hypoxia regulates the cystine glutamate exchanger (System Xc). ► Hypoxic preconditioning causes an increase in glutathione dependent on System Xc. ► Inhibition of system Xc attenuates hypoxic preconditioning neuroprotection.
