Optimisation of Fluorescent Fusion Protein Expression in Escherichia coli via Design of Experiments

Two derivates of Escherichia coli BL21 (DE3) were used for the expression of an IPTG-inducible fluorescent fusion protein ZsGreen-hEphB2-6His. The influence of cell specific growth rate, liquid phase temperature and IPTG inducer concentration on soluble and insoluble protein expression was studied. A multi-fermenter system BIOSTAT® Qplus-6 was configured for the execution of automated screening and optimisation experiments according to a Design of Experiments (DoE) strategy.
