In this paper, a novel optical probing strategy for the detection of amantadine has been successfully developed based on the competitive host–guest interaction of amantadine and rhodamine6G (R6G) with mono-[6-(2-aminoethylamino)-6-deoxy]-β-cyclodextin (EDA-CD). We synthesized the EDA-CD and graphene oxide (GO), and the EDA-CD functionalized graphene oxide (GO-CD) was prepared by covalently linking amino group of the EDA-CD with carboxylic acid groups on graphene oxide surface. R6G was chosen as a fluorescent probe because of its capability to enter into the cavity of EDA-CD. After R6G was incorporated into the hydrophobic inner cavity of GO-CD, significant fluorescence quenching was observed due to the fluorescence resonance energy transfer (FRET) between R6G and GO-CD complex. When amantadine was added to the GO-CD/R6G complex system, R6G would be released from the inner cavity of GO-CD and result in the fluorescence recovery due to the higher affinity between amantadine and EDA-CD, which could be used as a fluorescent probe for the detection of amantadine. The proposed method was successfully applied to the determination of amantadine in pharmaceutical capsule with good accuracy and satisfactory recovery.
