Peptidomimetics containing a vinyl ketone warhead as falcipain-2 inhibitors

► We report the synthesis of peptidomimetics with a vinyl ketone warhead. ► Their activity as falcipain-2 inhibitors have been evaluated. ► The realized structural modifications decreased the inhibitory activity. ► Docking studies allowed us to rationalize the activity profile of compounds.
