Synthesis of 4-(aminoalkyl) substituted 1,3-dioxanes as potent NMDA and σ receptor antagonists

► Side chain and ring homologues of dexoxadrol and etoxadrol were synthesized. ►
- 1,3-dioxanes with an aminoethyl residue are potent NMDA receptor antagonists. ► Whereas primary amines show high NMDA affinity, benzylamines represent potent σ1 ligands.
