A uniform human Wnt expression library reveals a shared secretory pathway and unique signaling activities

► We created a synchronized DNA plasmid set of all 19 human Wnts. ► Eighteen out of nineteen Wnts require PORCN-mediated palmitoylation to signal properly. ► All tested Wnts require WLS to signal through the β-catenin-dependent pathway. ► Fourteen of nineteen Wnts can activate β-catenin-dependent signaling. ► Wnts display significant variation in their stability, processing and secretion.
