• SC-CO2 was used for taxifolin (TAX) extraction from pinus bark.
• Response surface method was applied to optimize effective operating conditions.
• 32.13% TAX recovery was obtained from pine bark at optimum operating conditions.
• Developed model was validated by experimental TAX SFE data.

In this study, the extraction of taxifolin (TAX) from pine (pinus nigra) bark was evaluated by supercritical CO2 with ethanol as modifier and 30 min of static extraction time. Response surface methodology (RSM) was implemented using Minitab software 17 for design of experiments. The operating temperature (40–60 °C), pressure (10–30 MPa), dynamic extraction time (30–150 min) and CO2 flow rate (0.4–2 ml/min) were considered as the operating variables. The amount of extracted TAX was identified by high performance liquid chromatography (HPLC). Response surface analysis verified that R2 and adj.R2 of the model were 98.16% and 96.54%, respectively. The RSM predicted that the maximum extraction recovery of 34.13% was achieved at the optimal operating conditions to be the pressure of 19.3 MPa, temperature of 42.8∘C, CO2 flow rate of 1.9 ml/min and dynamic extraction time of 137.9 min. The accuracy of the modeling optimal TAX recovery was validated with triplicate experiments giving the average extraction recovery of 34 ± 2%.
