Analysis of Flavonoids and Phenolic Acids in Iris tectorum by HPLC-DAD-ESI-MSn

AimTo develop high performance liquid chromatography combined with photodiode-array detection and electrospray ionization multiple-stage mass spectrometry (HPLC-DAD-ESI-MSn) for the analysis and identification of flavonoids and phenolic acids in the rhizome of Iris tectorum Maxim‥MethodThe structures of flavonoids and phenolic acids were identified by chromatographic retention times, UV spectra as well as ESI-MSn spectra.ResultsTen isoflavones were identified as tectorigenin-7-O-β-glucosyl-4′-O-β-glucoside (3), tectoridin (5), iristectorin B (6), iristectorin A (7), iridin (8), genistein (11), tectorigenin (12), iristectorigenin A (14), iristectorigenin B (16), i and rigenin (17). Two flavanones, one flavonol and one flavanonol were tentatively identified as hesperetin (9), 5, 7, 3′-trihydroxy-6, 4′-dimethoxyflavanone (10), rhamnocitrin (13) and dihydrokaempferide (15), respectively. The three phenolic acids were tectoruside (1), androsin (2) and apocynin (4).ConclusionThe developed simple and rapid method is useful to rapidly identify the bioactive constituents in the rhizome of Iris tectorum. Two flavanones, hesperetin (9) and 5,7,3′-trihydroxy-6, 4′-dimethoxyflavanone (10) were identified from this species for the first time.

摘 要目的:建立高效液相色谱-电喷雾多级质谱法(HPLC-DAD-ESI-MSn), 鉴定鸢尾根茎中的黄酮和酚酸类成分。方法:利用样品的色谱的保留时间以及多级质谱分析, 对其中的黄酮和酚酸类成分的结构进行了鉴定。结果:十个黄酮分别鉴定为鸢尾苷元7-O-β-葡萄糖-4′-O-β-葡萄糖双糖苷 (3), 鸢尾苷 (5), 鸢尾新苷B (6), 鸢尾新苷 A (7), 野鸢尾苷 (8), 染料木素 (11), 鸢尾苷元 (12), 鸢尾甲黄素 A (14), 鸢尾甲黄素B (16) 和野鸢尾苷元 (17)。两个二氢黄酮, 一个黄酮醇和一个二氢黄酮醇暂时分别鉴定为橙皮素(9), 5, 7, 3′-三羟基-6, 4′-二甲氧基二氢黄酮 (10), 鼠李柠檬素 (13) 和二氢山奈酚 (15)。此外三个酚酸类成分为草夹竹桃双糖苷(1), 草夹竹桃苷 (2) 和夹竹桃麻素 (4)。结论:所建立的高效液相色谱-电喷雾多级质谱法可以用于快速鉴定鸢尾根茎中的活性成分。并首次在该植物中鉴定出橙皮素(9)、 5, 7, 3′-三羟基-6, 4′-二甲氧基二氢黄酮 (10)。
