Manipulation of the functions of hypoxia-inducible factors (HIFs), transcription factors that control the expression of multiple growth factors, is a potentially superior strategy for therapeutic angiogenesis than targeting a single angiogenic factor. In particular, HIF-2α plays an essential role in vascular remodeling and leads to mature angiogenesis by inducing the expression of selected angiogenic factors. HIF-2α is more stable than HIF-1α under mild hypoxia, a quality that may facilitate clinical application. The results of this animal study indicate that induction of HIF-2α in muscle cells is a possible strategy to promote perfusion recovery of ischemic injuries.
