Temperature dependence of interaction between double stranded DNA and Cy3 or Cy5

► We studied FRET between Cy3 and Cy 5 on dsDNA from 4 to 50 °C. ► The data was simulated according to the orientation-dependent model. ► Dyes are caped on the DNA duplex even at room temperature. ► Random motion makes more contribution at higher temperature.
