Pseudo half-molecules of the ABC transporter, COMATOSE, bind Pex19 and target to peroxisomes independently but are both required for activity

► CTS pseudo-half molecules target independently to the peroxisome membrane in yeast. ► Both half molecules bind yeast and plant PEX19 in vitro. ► Arabidopsis PEX19 isoforms do not complement the yeast pex19Δ mutant. ► N- or C-terminal half molecules are not functional when expressed alone in yeast. ► Co-expression of both half molecules restores function comparable to full-length CTS.
