A facile no-carrier-added one pot micro-molar scale radiolabelled synthesis of 2-[14C]–uracil from [14C]–urea and propiolic acid in the presence of polyphosphoric acid (PPA) with an yield of 48.33% and with radiochemical purity of 98% is reported in this paper.

► The method adopted by us has not been reported so far for the 2-[14C]–uracil synthesis, as far as we know.
► Our aim to obtain a high specific activity product having high radiochemical purity met with success.
► To achieve the above, the radiosynthesis was carried out on micromolar scale.
► A radiochemical yield of 48.33% with radiochemical purity of more than 98% has been realized.
► The use of fuming sulphuric acid in the reaction was avoided to prevent any degradation of product.
