Glucokinase activation – a strategy for T2D therapy: recent developments

Glucokinase (GK) plays a key role in glucose homeostasis primarily through its modulatory actions in the pancreatic β-cells and the liver. There has been a growing interest over the past few years in discovery and development of GK activators (GKAs), as a novel therapy to treat type 2 diabetes (T2D). In this review, recent publications on GKAs that reported full characterization of lead compounds and current hypothesis on mechanism of GK activation will be discussed.
