En los últimos años se ha identificado gran cantidad de mutaciones y polimorfismos en pacientes afectados de enfermedades neurodegenerativas. El descubrimiento de estas alteraciones genéticas ha ayudado a comprender mejor la patogenia de estas enfermedades. En la presente revisión se exponen las alteraciones genéticas halladas hasta la actualidad que determinan o predisponen a experimentar diferentes enfermedades neurodegenerativas, como son la enfermedad de Huntington, la enfermedad de Alzheimer, la demencia frontotemporal, la enfermedad de Parkinson y otros parkinsonismos. Asimismo, se comenta el posible mecanismo patogénico de estas mutaciones, sus características clínicas principales y la utilidad de esta información en el diagnóstico y el tratamiento de estas enfermedades.

A large number of mutations and polymorphisms associated with neurodegenerative disorders have been described during the last years. These findings have been helpful to improve our knowledge about the pathogenesis of these disorders. In this review we describe the genetic alterations and variants that cause or predispose to develop several neurodegenerative disorders, such as Huntington's disease, Alzheimer's disease, frontotemporal dementia, Parkinson's disease and other parkinsonisms. We also comment on the possible pathogenic mechanism of these mutations, clinical features and the usefulness of this information for the diagnosis and management of these disorders.
