► Transmeningeal acetylcholine (Ach) induces seizures in the rat neocortex. ► Transmeningeal muscimol can prevent Ach-induced neocortical seizures in rats. ► Daily transmeningeal muscimol maintains its antiepileptic efficacy for 3 weeks. ► Periodic transmeningeal muscimol does not induce tolerance for 3 weeks.
