Marker-assisted selection for two dominant powdery mildew resistance genes introgressed into a hybrid grape population

▶ MAS was applied to track the inheritance of Run1 and Ren1 (R) genes in a BC5 grape hybrid. ▶ Run1 was introgressed from a Muscadinia rotundifolia × Vitis vinifera BC4 hybrid. ▶ Ren1 was introgressed from the PM-resistant V. vinifera L. variety Kishmish vatkana. ▶ We applied SSR and BAC-derived CB markers to follow the resistance genes in a BC5 progeny. ▶ The PM resistance co-segregated with the presence of at least one R locus-linked marker. ▶ We developed a multiplex PCR and agarose gel based method to detect both Run1 and Ren1 in one step. ▶ MAS offers an accurate method to select genotypes carrying multiple loci of interest.
