An off-line mode coupled supercritical fluid extraction (SFE) with high-performance liquid chromatography and electro-spray ion trap mass spectrometry (HPLC-ESI-MS) analytical method was applied to analyze the chemical components of Ganoderma sinense (G. sinense). The ultra-fine powder of fruit-bodies of G. sinense was extracted by SFE and a HPLC-ESI-MS method was established for simultaneously determining chemical components of G. sinense, and the total extraction yield was 3.75% (w/w). The method of combining diode-array detection (DAD) with MS, was successfully used to detect the chemical components of G. sinense. Most of the peaks exhibited maximum absorptions at 280 and 210Â nm, especially at 280Â nm. About 20 peaks were acquired in positive and negative ion detection mode, and were analyzed by electro-spray ion trap mass spectrometry (ESI-ITMS). The results indicated that the molecular weight distribution (m/z: 188-614) of main compounds was identified according to the molecular ions in MS spectra.
