Nanogrooves smaller than the width of neurites were created with interference lithography on Polyethylene terephthalate (PET) substrates. PC12 cells were grown and stimulated, and the neurites were shown to be directed by the nano-topology of the surface. We have shown with directed neurites, 3 miRNAs were regulated differentially compared to cells with randomly grown neurites. By disrupting the regulations of miRNAs, the neurite directing effect was reduced. The finding shows involvement of miRNAs in the process of neurite directing, implies significant involvement of miRNAs in regulating intracellular processes with extracellular mechanical cues.137
