آشنایی با موضوع

پپسین مهم ترین آنزیم سلول های اصلی معده است. این آنزیم بصورت پپسینوژن که فاقد فعالیت گوارشی است در داخل سلول ها ساخته می شود. ولی این ماده با ورود به مجرای غدد در حضور اسید کلریدریک بلافاصله فعال شده و بشکل پپسین در می‎آید. پپسین در محیط هایی که اسیدیته زیادی دارند آنزیم پروتئولیتیک بسیار فعالی است. اما اگر PH محیط از 5 تجاوز نماید، اثر پروتئولیتیک پپسین بسیار ضعیف شده و ممکن است کاملاً غیرفعال گردد. از این رو در عمل گوارشی معدی اسید کلریدریک به اندازه پپسین ضروری است. پپسین توسط فیزیولوژیست آلمانی تئودور شوان در سال 1836 کشف شد. پپسین در محیط با اسیدیته بالا آنزیم پروتئولیتیک بسیار فعالی است و بخصوص پیوند بین اسیدآمینه های آروماتیک و هیدروفوبیک را میشکند. اما اگر PH محیط از 5 تجاوز نماید (با ورود غذا به روده)، اثر پروتئولیتیک پپسین بسیار ضعیف شده و ممکن است کاملاً غیرفعال گردد. پپسین فقط در هضم پروتئینها نقش دارد و در هضم کربوهیدرات و چربی تاثیری ندارد. یکی از کاربردهای پپسین در صنایع غذایی است که از آنزیم پپسین برای دلمه کردن شیر در تولید پنیر استفاده می‌شود. از آنزیم پپسین همچنین ممکن است برای فرآوری پروتئین‌ سویا و ژلاتین استفاده شود. از این آنزیم ممکن است برای تهیه مواد هیدرولیز شده از پروتئین حیوانی و گیاهی به عنوان طعم دهنده در مواد غذایی یا نوشیدنی‌ها استفاده شود. از آنزیم پپسین می‌توان برای ارزیابی قابلیت هضم پروتئین‌ها استفاده کرد. در صنعت چرم، از پپسین برای حذف بافت و موها و نرم کردن چرم استفاده می‌شود. در صنعت آزمایشگاهی نیز از آنزیم پپسین برای آنالیز پروتئین‌های مختلف استفاده می‌شود.
