Development of a novel orthotopic non-small cell lung cancer model and therapeutic benefit of 2′-(2-bromohexadecanoyl)-docetaxel conjugate nanoparticles

A novel orthotopic non-small cell lung cancer model was established in nude mice with luciferase-expressing A549 cells. The model was characterized by a survival study, histological staining, bioluminescence imaging and micro PET/CT imaging. Intravenous administration of lipid-based nanoparticles containing 2′-(2-bromohexadecanoyl)-docetaxel conjugate significantly prolonged progression-free survival and overall median survival as compared to Taxotere®. Whole-body bioluminescence imaging provided reliable signals for both untreated tumors and relapsed tumors.130
