Improvement of σ1 receptor affinity by late-stage C-H-bond arylation of spirocyclic lactones

Selective α- and β-arylation of complex thiophene annulated lactones with iodoarenes in the last step of the synthesis allows fine tuning of σ1 receptor affinity.
