Structure-based design, synthesis, and biological evaluation of dihydroquinazoline-derived potent β-secretase inhibitors

β-Secretase inhibitor 4f showed an enzyme Ki of 13 nM and a cellular IC50 value of 21 nM. A molecular model of 4f provided insight into the ligand-binding site interactions.
