► Na+-K+ pump function is critical for regulation of intracellular Na+ and Ca2+. ► The Na+-K+ pump is inhibited by glutathionylation of its β1 subunit at Cys46. ► NADPH oxidase mediates inhibition of the cardiac Na+-K+ pump. ► Endothelial NO synthase mediates stimulation of the cardiac Na+-K+ pump. ► Approaches to reverse oxidative Na+-K+ pump inhibition may be cardioprotective.
