► Asymmetric liposome particles (ALPs) with highly efficient siRNA encapsulation and negatively charged surface. ► The ALPs protect the encapsulated siRNA from RNase A. ► The cellular uptake of ALPs is only induced by attachment of ligands interacting with cell surface receptors. ► The internalized siRNA successfully inhibits the target gene expression.
