► Transporters are mandatory for proper thyroid hormone (TH) metabolism and action. ► MCT8 represents a highly specific TH transporter. ► Mutations in MCT8 are linked to neurological symptoms and abnormal TH profile. ► We review the consequences of Mct8 deficiency in mice. ► We report on the role of other TH transporters such as Oatp1c1 in mice.
