► The crystal structure of DwL reveals important differences which can explain why DwL do not present anti-inflammatory effects. ► Differences in interaction patterns between the X-Man and LEU99 are due to differences in the LEU99 side chain conformation. ► DwL differs structurally from other Dioclea lectins with regard to the conformation of CRD and related biological activities.
