► GSTO catalyze deglutathionylation of aromatic molecules and possess esterase activity. ► These activities are depending on a conserved catalytic cysteine. ► This feature has been used to develop a specific substrate screening method. ► We identified tetralone and terpenes as specific substrate/ligands of PcGSTOs.
