► NO and CTS effects on the stretch response of the frog heart have been evaluated. ► CTS improves the sensitivity to preload increases similar to that exerted by NO. ► CTS action occurs via EE-induced NO release involving a PI3K-NOS-cGMP pathway. ► CTS acts as an endogenous NOS activator.
