Modeling chronic olanzapine exposure using osmotic minipumps: Pharmacological limitations

► Antipsychotics administered via osmotic minipump model chronic treatment in humans. ► We examined olanzapine administered over 1 month via osmotic minipump in rats. ► Results indicated a monophasic exponential decay and half-life of 14.8 days. ► Oxidation of olanzapine appears to be involved. ► The administration interval for olanzapine should be confined to 2 weeks.
