Production of recombinant Rhizopus oryzae lipase by the yeast Yarrowia lipolytica results in increased enzymatic thermostability

►Rhizopus oryzae lipase (ROL) was expressed in the yeast Yarrowia lipolytica. ► The combination of pre- and prosequences for ROL secretion was tested. ► Lipase production was highest (7.6 U/mL) with a hybrid variant of preAEP and proROL. ► The most active clone was used for enzyme purification and characterization. ► Recombinant ROL exhibited higher thermostability than previously reported.
