Preferential base pairing modes of T·T mismatches

► We study the sequence context effect on the pairing modes of T·T mismatches. ► We demonstrate the presence of preferential pairing mode in T·T mismatches. ► The pairing mode with flanking purines differs from that with flanking pyrimidines. ► These different pairing modes usually undergo exchange at room temperature.
