► Sulfatases play an important role in alveolar formation and lung function. ► We identified that over accumulation of sulfated glycosaminoglycans leads to upregulation of TGFβ signaling. ► Anti-TGFβ therapy leads to normal lung development and function in Sumf1−/− mice.
